Unliganded thyroid hormone receptor 1 impairs adult hippocampal neurogenesis

چکیده
Thyroid hormone regulates adult hippocampal neurogenesis, a process involved in key functions, such as learning, memory, and mood regulation. We addressed the role of thyroid hormone receptor TR 1 in adult hippocampal neurogenesis, using mice harboring a TR 1 null allele (TR 1 / ), overexpressing TR 1 6-fold (TR 2 / ), and a mutant TR 1 (TR 1 ) with a 10-fold lower affinity to the ligand. While hippocampal progenitor proliferation was unaltered, TR 1 / mice exhibited a significant increase in doublecortin-positive immature neurons and increased survival of bromodeoxyuridine-positive (BrdU ) progenitors as compared to wild-type controls. In contrast, the TR 1 /m and the TR 2 / mice, where the overexpressed TR 1 acts as an aporeceptor, showed a significant decline in surviving BrdU progenitors. TR 1 / and TR 2 / mice showed opposing effects on neurogenic markers like polysialylated neural cell adhesion molecule and stathmin. The decreased progenitor survival in the TR 2 / and TR 1 /m mice could be rescued by thyroid hormone treatment, as was the decline in neuronal differentiation seen in the TR 1 /m mice. These mice also exhibited a decrease in NeuroD cell numbers in the dentate gyrus, suggesting an effect on early postmitotic progenitors. Our results provide the first evidence of a role for unliganded TR 1 in modulating the deleterious effects of hypothyroidism on adult hippocampal neurogenesis.— Kapoor, R., van Hogerlinden, M., Wallis, K., Ghosh, H., Nordstrom, K., Vennstrom, B., Vaidya, V. Thyroid hormone regulates hippocampal neurogenesis in the adult rat brain.
We have examined the influence of thyroid hormone on adult hippocampal neurogenesis, which encompasses the proliferation, survival and differentiation of dentate granule cell progenitors. Using bromodeoxyuridine (BrdU), we demonstrate that adult-onset hypothyroidism significantly decreases hippocampal neurogenesis. This decline is predominantly the consequence of a significant decrease in the s...

The transcriptional properties of unliganded thyroid hormone receptors are thought to cause the misdevelopment during hypothyroidism of several functions essential for adult life. To specifically determine the role of unliganded thyroid hormone receptor alpha1 (TRalpha1) in neuronal tissues, we introduced a mutation into the mouse TRalpha1 gene that lowers affinity to thyroid hormone (TH) 10-fo...

Disrupted thyroid hormone function evokes severe physiological consequences in the immature brain. In adulthood, although clinical reports document an effect of thyroid hormone status on mood and cognition, the molecular and cellular changes underlying these behavioural effects are poorly understood. More recently, the subtle effects of thyroid hormone on structural plasticity in the mature bra...
متن کاملRestricted CD4+ T cell receptor repertoire impairs cognitive function via alteration of Th2 cytokine levels
Despite the effects of CD4+ T cell dysfunction on cognitive and behavioral impairment are well established, the effects of Th2 cytokines on the adult hippocampal neurogenesis and cognitive function in restricted CD4+ T cell receptor (TCR) repertoire model have not been fully elucidate. We found that mice with restricted CD4+ repertoire TCR showed decreased adult hippocampal neurogenesis using O...

Neurogenesis occurs throughout life in several regions of the brain. In this lecture, a new sight for the role of the dentate gyrus and adult hippocampal neurogenesis in anxiety disorders will be discussed. The region that has obtained the most attention for its involvement in the neurogenesis of affective and anxiety disorders are the hippocampal and dentate gyrus. Evidence strongly suggests t...
